    Description of Improving Previously Existing Parts/Devices

    In our project, we have done improvement of the function/characterization on two previously existing BioBrick Part/Device(BBa_I721002 and BBa_K1613017)

    1.BBa_I721002

    (1).Design of Circuit

    It is a lead binding protein (PbrR), belonging to a MerR family. It plays a role of regulator in gene transcription of lead resistance operon. In order to measure the generation of PbrR, we designed two types of PbrR genetic circuits to measure the PbR according to the GFP expression downstream.

    (a)Autoregulated PbrR circuit(BBa_K1961004): an autoregulated generator of PbrR measured by GFP expression downstream. PbrR and GFP expression is under the control of TetR and promoter PTet (Figure 1(a))

    (b)Constitutive PbrR circuit(BBa_K1961005): a constitutive generator of PbrR measured by GFP expression downstream. PbrR and GFP expression is under the constitutive promoter (Pcon, BBa_J23102 ) (Figure 1(b))

    In order to investigate the response of PbrR in these two circuits, we used GFP as a reporter downstream as a proxy to measure the PbrR produced by two circuits. For autoregulation PbrR circuit, we want to study in detail about the response of PbrR protein with increasing amount of inducer tetracylcine. The advantage of this autoregulation circuit is to get very precisely gradual response to the inducer, which implies that the amount PbrR protein will be a smooth function of inducer. In contrast, we also designed the constitutive PbrR generator. This circuit enables us to measure the background expression of PbrR in E. Coli. This will help us to compare and recognize the generation of PbrR under autoregulation. All the constructs used here, the Hill coefficient will be calculated to disclose the intrinsic property of genetic circuits and be a guide for our biosensor project.

    BBa_K1613017

    This was a previously existing composite part can detect lead ions. In 2015, HSNU-TAIPEI didn’t succeed and didn’t ship this part. This year, we re-do the ligation of this composite part as new part (BBa_K1961006) and tested its fluorescence.
