Notebook

Wednesday, September 21st

Tasks:

Sam & Tyler

  • Checked pET28a-Cas9 and pET28a-Cas9-SS Gibson plates
    • pick for overnight cultures
  • Checked pSB1C3-Cas9 ligation plates
    • pick for overnight cultures
  • Check overnight cultures from Tuesday (gRNA + Cas9 cotransformation)
    • Didn’t grow
    • Made two more from the plate with lots of colonies
    • Other plate only has one colony left so we’re growing it up on the plate in the incubator for now
