BioBricks
In the process of designing our system, we have created seven BioBricks and further characterised an existing one.
We decided to further characterise the BioBrick osmY, which was already present in the iGEM registry thanks to the 2012 Washington team. To characterise this brick, we attached a HA tag. The HA tag is in place for us to know whether or not our model cell E. coli is secreting the protein osmY.
The first two BioBricks we designed were Hfq-Serratia and Hfq-E.coli. Hfq is an RNA binding protein, which we have then further characterised by attaching our sRNA expression system in order to further develop our system and create two more bricks: sRNA-Hfq-E. coli and sRNA-Hfq-Serratia.
In the process of making our expression system, we have also designed the BioBricks osmY-Hfq-E. coli and osmY-Hfq-Serratia which have been added to the registry and will hopefully be of use to future teams.
