  • Checked new overnight culture from the one plate with lots of colonies
    • Still didn’t grow much
    • Made a new overnight culture from the other plate once the colony is big enough
  • Miniprepped pET28a-Cas9 and pET28a-Cas9-SS Gibson cultures (leave 500uL culture for glycerol stocks)
    • Accidentally used all of the culture
  • Miniprepped pSB1C3-Cas9 ligation cultures (leave 500uL culture for glycerol stocks)
    • Accidentally used all of the culture
