Tasks:

Jordan

  • DpnI digest of SS-Cas9 PCR

Paul

  • PCR of all SS-Cas9 with new primers

Sara

  • Ran a gel screen on Cas9 and SS with restriction digest ends for assembly into pSB1C3 plasmid
  • It did not work

Tasfia

  • Retransform all pET28a Parts (edit: just use colonies from last time, using a combination of colony pcr and restriction digest to figure out which colonies match up with which sequencing prep)
  • Grow up cultures of pET28a parts

Tyler

  • Pick Up Primers
  • Take cas9+gRNA cultures out of shaking incubator
