Our project centers on solving a problem many synthetic biologists face known as supercoiling, which greatly affects the predictability of gene expression. In order to solve this problem, we have designed a plasmid where two genes—RFP and GFP—are constructed onto a plasmid. In between them, a “spacer” part can be added between the genes. Here, either a set number of base pairs can be added or a clamp site for cas9 can be added to test whether or not supercoiling can be lessened. We utilize Golden Gate Assembly to help us easily build many variations of these plasmids by cloning with different parts.
