Meeting Minutes

  1. Project description due next Friday (1st July 2016)

  2. Remaining deadlines added to calendar

  3. Booking to be started for IWDBA conference – 4 students will go, the remaining 4 will go to the UK iGEM meetup

  4. Discussed issues with interlab study – left reader for 9 hours, should be 100ml of sample removed every hour and placed on ice for 6 hours
    - more FITC should be ordered, standard curve should be plotted and readings compared with other teams

  5. Lightbulb designs – structure of 3 devices discussed with mechanisms of regulation and expression – use different strength promoters?.
Obtain quotes from DNA 2.0 and other services for the cost of synthesis
Increased GFP output associated with use of repeated sigma factors
Use of multiple stop codons to ensure proper dissociation

  6. Modification of designs
-Use of superfolder GFP for brighter fluorescence
-Potentially weak RBS for not overloading system
-Model oscillatory behavior of stress response
-Examine adverse effects of sustained stress response on growth
    -Optimal spacing between codons
-Check degradation GFP tags – proteases cause quicker fluorescence decay

  7. Microbial fuel cells – potential application to trickle charge capacitor
Obtained parts & chemicals ordered for ChemEng cell kit
Use of mixed geobacter and Shewanella cultures?

  8. Contact Jennifer Hallinan at Macquarie University

  9. Survey appropriate age groups – secondary schools?
Check for approved samples of ethics forms
Lack of modelling in secondary schools
Potentially compare data mined responses to survey results
    Engage with teachers (PARTNERS?)

  10. Culture shock – ‘fusion of two paradigms’

  11. Go over medal requirements for bronze – theory only, does not need to actually work

  12. Acquire electronics – multimeter / electrodes for cell

  13. Contact breadboard manufacturers

  14. Contact 3D manufacture locations – MakerSpace, Fablab
