Meeting Minutes

The minutes from the previous meeting were unanimously approved.

We recapped the upcoming the deadlines.

Team banners are due on the 7th of October

Team banner was mocked up and is present on the google drive. Some suggestions were made how to improve the banner including smaller logo and text and a picture that illustrates our project.

Jem suggested that design of the banner would be like Duracell battery and a mock up can be found on igem general mailing list.

We reported back on the individual sub-teams.

Simulation

The last level of thought experiment requires more work than expected so it was decided to consult Jake about scrapping it off.

The experiment should be demonstrated to team supervisors and PEALS to get feedback from experts and non experts.

The video will be sent to Edinburgh team and we expect a video from them as well.

Wiki

Rupert has made a list of empty wiki pages and assigned people or they self-assigned to fill the pages.

It was suggested that each section would have an active link which says "For Judges:click here".

Motivation section of front page is very important. It should be like an abstract that contains short project description, results, etc. that would make people to understand our project without going anywhere else on the wiki.

Light Bulb/Battery

Last week was spent getting to the right gain value of the experiment because it was always maxing out. This week we were running experiments with IDT and DNA 2.0 constructs, positive and negative control and BL21 backbone at 42 degrees using plate reader. It was noticed that after heat shock at 42 degrees we get more fluorescence than without.

The next experiment suggested was to grow cultures at 37 degrees in duplicates, then take some of them and heat shock. Then leave some of heat shocked ones to grow at 42 degrees and place the other half back to 37 degrees to see the difference it makes.

It was noted that all experiments should be done by the end of next week.

We reflected on our progress with respect to the registry.

Registry is the most important part of the project so it needs sorting out. Lauren and Martin will go through the registry on Monday to add all the missing parts.

Each composite part should consist of basic parts that also have an entry.

rpoH sequence is missing from light bulb construct and needs to be added before creating a composite part.

Parts should be sent to iGEM HQ by the end of next week with FEDEX (action point). They are already prepared in Devonshire.

All parts need design and characterization in the registry.

Ollie, Kerry and Rupert will present at the Jamboree, with Josh and Emilija as reserves.

Ollie is writing/editing the script and hopefully it will be ready by the next meeting.

Use the same poster template, just edit text and add characterization of data.

T-shirts will need to be ordered soon.

Cream and burgundy are suggested colours to match the colour of the wiki page. Design should be done by next meeting.
