October 3rd 2016
This week we planned to run the plate reader experiments with much more success than we have had previously. We inoculated fresh cultures of all the samples to be tested.

Today was the first day of the next attempt at the plate reader experiment this time using a gain value of 1000, but continued with the same protocol described earlier. We first ran the plate at a temperature of 30°C. We inoculated fresh LB culture in order to continue with the experiment tomorrow.

We checked our data that had been collected over night and the results were extremely promising. We had much lower ‘noise’ and the gain value appeared much more appropriate. Our results can be seen here. We again set up the plate reader but this time at a temperature of 37°C. We inoculated fresh LB culture in order to continue with the experiment tomorrow.

We again collected our data from the overnight plate reader at 37°C and again the results were as expected, we set up the final plate reader run at 42°C. Our results can be seen here. We inoculated fresh LB culture in order to continue with the experiment tomorrow.

We collected the data from the final run in the plate reader and our constructs appeared to have worked exactly as expected and the results can be seen here. Next week we plan to replicate the Stanford experiment to further characterize our parts with regards to electrical current.
