In the morning we checked the plate reader from the previous day. After looking at the results, we concluded that the gain of 100 was too low for the fluorescence, as the positive control was saturated after 4 hours, and our construct was saturated after 8 hours. We carried out the same plate reader protocol again, however, this time we set the gain to 0 and allow the plate to run for a full 24 hours instead of 22 hours.
